Description
Dynabeads Streptavidin for In Vitro Transcription offers an innovative technology for simple, flexible, and automation-ready mRNA synthesis in scales ranging from microgram to gram. The 1-μm monosized, monodispersed, paramagnetic beads are covalently coupled to streptavidin molecules, ready for binding to a biotinylated DNA template. The workflow is designed to minimize the effort and cost of plasmid preparation while enabling storage and reuse of bead-template complex in a successive solid-phase in vitro transcription (IVT) reaction.

Reusable bead-based IVT
The biotinylated template is made by PCR amplification of the target sequence in a plasmid or a synthetic DNA construct. This uses a biotinylated forward primer, located 30-100 base pairs upstream of the T7-promoter, and a non-biotinylated reverse primer. As an alternative to PCR amplification, a linearized plasmid can be biotinylated by a fill-in reaction of biotin-dUTP in the 5' overhang sequence. The biotinylated template is then immobilized directly to Dynabeads Streptavidin for IVT without the need for prior purification. The bead-bound template is used directly in the IVT, then removed from the synthesized mRNA by magnetic separation. The template can then be reused at least six times in consecutive IVT reactions. The protocol is scalable from micrograms to grams of mRNA.
Automation ready with the KingFisher Purification System
The 1-μm Dynabeads magnetic beads are ideal for high-throughput enrichment. This process can be automated using any of the KingFisher Sample Purification Systems or liquid handlers.
